The purpose of this study was characterized by ISSR markers some Spanish olive cultivars marketed in Chile, among them a Chilean cultivar of economic importance named Sevillana (Azapa). A modified and suitable CTAB protocol for DNA extraction for drupes and leaves tissue, without the use of liquid nitrogen was evaluated. Seven olive (Olea europaea L.) cultivars from INIA Germplasm Bank, located in Atacama Region, were screened using inter-simple sequence repeats (ISSRs) markers. Fifteen selected ISSR primers that produced 153 reproducible amplification bands and 130 polymorphic bands (87%) were evaluated. The combinations of nine ISSR primers were able to distinguish all cultivars in study. With UBC807, UBC814 and UBC850 primers we detect three exclusive bands in Sevillana cultivar. Phylogenetic analysis relationship shows that there is a greater closeness between Sevillana and Picuda local cultivars instead the spanish origin Gordal Sevillana. In this study, a molecular identification method is proposed in order to discriminate Sevillana of other table olive cultivars that are marketed in the country.
